August 12th, 2008. Malignant mesothelioma in garment sewing-machine workers

Conclusion: The results of this investigation suggest assigning at least "possible occupational asbestos exposure" to those cases employed in clothing manufacture since the 1950's, who used sewing-machines or pressing machines, according to the Re.Na.M guidelines. Other possible occupational exposures to asbestos in this working sector cannot be excluded; when the simple interview of patients does not reveal such exposures further investigations are needed in order to demonstrate all the possible circumstances of exposure.

August 9th, 2008. Clinical Activity of Vinflunine in Transitional Cell Carcinoma of the Urothelium and Other Solid Tumors

The activity and tolerability of this agent warrant further investigation. Phase 3 trials are underway to further define the extent of clinical benefit provided by vinflunine in patients with advanced solid malignancies.

August 9th, 2008. Mesothelioma mortality in Brazil, 1980-2003

Mortality was highest among males and people over age 65. Given the history of asbestos exposure in Brazil, our findings support the need for policies that limit or ban the use of this product.

August 8th, 2008. A Meta-Analysis of Asbestos-Related Cancer Risk That Addresses Fiber Size and Mineral Type

(2007) recently analyzed by TEM archived air samples from the South Carolina plant to determine a detailed distribution of fiber lengths up to lengths of 40 μ m and greater. If similar data become available for Quebec, perhaps these two size distributions can be used to eliminate the discrepancy between these two studies.

August 8th, 2008. Sensitivity analysis of the relationship between disease occurrence and distance from a putative source of pollution

However, as the models appeared to be sensitive to modeling choices, we suggest that any analysis of disease risk around a putative source should be integrated with a careful sensitivity analysis and possibly with prior knowledge. The choice of prior distribution is extremely important and should be based on epidemiological considerations.

August 8th, 2008. Novel Functional View of the Crocidolite Asbestos-Treated A549 Human Lung Epithelial Transcriptome Reveals an Intricate Network of Pathways with Opposing Functions

Conclusions: Our analyses demonstrate the power of combining a statistically robust, comprehensive dataset and a functional network genomics approach to 1) identify and explore relationships between genes of known importance 2) identify novel candidate genes, and 3) observe the complex interplay between genes/gene products that function in seemingly different processes. This study represents the first function-based global approach toward understanding the response of human lung epithelial cells to the carcinogen crocidolite. Importantly, our investigation paints a much broader landscape for the crocidolite response than was previously appreciated and reveals novel paths to study. Our graphical representations of the function-based global network will be a valuable resource to model new research findings.

August 7th, 2008. The serine protease HtrA1 is a novel prognostic factor for human mesothelioma

Conclusion: This is the first study of the relationship between HtrA1 expression and survival of mesothelioma patients. The data obtained strongly indicate the utilization of HtrA1 expression as a prognostic parameter for mesothelioma and suggest this serine protease as a possible molecular target for the treatment of malignant mesotheliomas.

Conclusions: HDRs are useful in addition, but not in substitution, to active search of MM cases.

August 6th, 2008. Incidence of malignant mesothelioma in Piedmont, 1990-2001

Conclusions: the rise in incidence might be due to increased diagnostic awareness for MM. An analysis by birth-cohort, however, revealed a cohort-effect, therefore it is unlikely that diagnostic bias alone could explain the observed trend. Among men residing at diagnosis in the province of Turin, but not among women nor in both sexes outside Turin province, the increase in incidence has levelled off in the most recent calendar period; for them exposures stopped on average seven years earlier than those of residents in other areas of Piedmont.

Herein, we review the biochemical characteristics of RI ribonuclease complexes and progress towards the development of mammalian ribonuclease-based chemotherapeutics through the elicitation of RI-evasion.

August 5th, 2008. Onconase and Amphinase, the Antitumor Ribonucleases from Rana pipiens Oocytes

Onconase and Amphinase are highly cationic molecules and their preferential toxicity towards cancer cells (having distinctly higher negative charge compared to normal cells) may depend on increased binding efficiency to the cell surface by electrostatic interactions. Here we will discuss the structures of Onconase and Amphinase and the molecular basis for their enzymatic and anticancer functions.

August 5th, 2008. Antibody-onconase conjugates: cytotoxicity and intracellular routing

Antibody-onconase conjugates are effective in preclinical models, cause little non-specific toxicities in mice and have favorable formulation properties. Understanding the reason for their potency coupled with understanding novel RNA-based mechanisms of tumor cell death will lead to improved variations of targeted onconase.

Conjugation of RNases to internalizing tumor-targeting monoclonal antibodies was shown to enhance specific cell killing by several orders of magnitude both in vitro and in animal models. In this review we describe the development and current status of genetically engineered 2(nd) generation immunoRNases as promising novel anti-cancer therapeutics.

Conclusion: Serum mesothelin remains the most specific marker for the diagnosis of mesothelioma.

These drugs function at a variety of steps within the HGF-MET pathway, including MET expression at the RNA or protein level, the ligand-receptor interaction, and tyrosine kinase function. This paper will review the structure, function, mechanisms of tumorigenesis, and potential for therapeutic inhibition of the MET receptor in lung cancer and mesothelioma.
