June 30th, 2007. Malignant mesothelioma of the pleura in a truck driver

Results: The subject had worked for more than 30 years as a truck driver operating on long distances on truck models described in literature as contaminated by asbestos fibres. He had not transported materials made of asbestos, and had not carried out maintenance on the trucks, nor had he any non-occupational sources of exposure to asbestos. Thus the mesothelioma was related to occupational exposure and procedures were initiated for reporting a suspected occupational disease.

June 28th, 2007. Caffeine markedly sensitizes human mesothelioma cell lines to pemetrexed

Pemetrexed sensitization by caffeine was associated with an increase in pemetrexed-induced phosphorylation of ataxia-telangiectasia-mutated (ATM) and Chk1. These data indicate that caffeine and its analog, theobromine, may be a useful approach to enhance pemetrexed-based chemotherapy.

June 26th, 2007. A randomised controlled trial of intervention site radiotherapy in malignant pleural mesothelioma

Conclusions: Prophylactic drain site radiotherapy in malignant pleural mesothelioma does not reduce the incidence of tumour seeding by the margin indicated by previous studies.

June 26th, 2007. Peritoneal mesothelioma: an inusual clinical presentation in a patient without exposure to asbestos

The patient received treatment with systemic palliative chemotherapy, cisplatin-pemetrexed. After three cycles, partial response was observed, but the evolution was fatal due to secondary toxicity of chemotherapy.

June 23rd, 2007. Management of malignant pleural effusion

PET imaging could be of major interest in the diagnosis of mesothelioma and prognosis evaluation. Mesothelioma prognosis has recently been modified by the association of cisplatin with pemetrexed, a new antimetabolite agent, but mesothelioma remains seldom curable.

June 22nd, 2007. The Diagnostic Role of Claudins in Serous Effusions

Our data suggest that expression of claudin-3 or claudin-7 is specific for adenocarcinoma and rules out the diagnosis of cells as mesothelial and that absence of claudin-1 expression excludes ovarian carcinoma as the possible origin of metastatic adenocarcinoma. Claudins may, therefore, be of diagnostic value in effusion cytology.

June 22nd, 2007. Control of human mesothelin-expressing tumors by DNA vaccines

Furthermore, we found CD4+ and CD8+ T-cell immune responses as well as the humoral immune responses are important for the observed antitumor effects in vaccinated mice. Our data indicated that vaccination with DNA vaccine targeting Hmeso could generate potent antitumor effects against mesothelin-expressing tumors through both T cell-mediated immunity as well as antibody-mediated immunity.

June 19th, 2007. A procedure for the isolation of asbestos bodies from lung tissue by exploiting their magnetic properties: a new approach to asbestos body study

Magnetically isolated AB were shown to be cytotoxic and to activate free radical production from inflammatory cells at a higher extent than that induced by bodies obtained by chemical digestion. The finding that hypochlorite-treated AB induce DNA damage, while AB obtained by the method described in this article failed to do so, together with the differential behavior of these bodies toward inflammatory cells, suggests that native asbestos bodies should be used to investigate the pathogenetic role of these structures.

June 19th, 2007. Mesothelioma mortality in Greece from 1983 to 2003

Conclusion: Our data suggest a change in the balance of risk away from traditional asbestos exposure industries to industries where one could describe the exposure as secondary such as plumbers, technicians, drivers, farmers. Also, we found out that the higher cause-specific mortality rate was 0.38/100,000 population in Epirus, the lower was 0.025/100,000 in Thessalia and the national average rate was 0.10/100,000 population.

June 19th, 2007. The essential role of the mitochondria and reactive oxygen species in Cisplatin-mediated enhancement of fas ligand-induced apoptosis in malignant pleural mesothelioma

Our data indicate that the mitochondria-dependent feedback loop of the caspase activation cascade and the generation of ROS are both essential in mediating profound cytotoxicity and apoptosis of MPM cells treated with CDDP and sFasL. This mechanistic study establishes a the translational framework for the clinical application of sequential CDDP/sFasL in the treatment of MPM.

June 19th, 2007. Establishment and characterization of two distinct malignant mesothelioma cell lines with common clonal origin

Remarkably, one xenograft from MM-Z03E revealed overexpression of p53 and widely invasive growth. In conclusion, both cell lines are useful in vivo and in vitro model systems to study the underlying genetic mechanisms of biphasic differentiation in MM, which can be of certain value considering the increasing relevance of assessing MM tumor biology for the clinical management of this disease.

June 19th, 2007. Risk of developing mesothelioma due to neighborhood exposure to asbestos

As a whole, relative risk (RR) of neighborhood exposure in crocidolite and amosite mines was about 10 to 30 and RR in major asbestos factories was about 5 to 20. On the other hand, statistically significant RR of neighborhood exposure was not observed in chrysotile mines and some asbestos facilities.

June 15th, 2007. Short hydration regimen and nephrotoxicity of intermediate to high-dose cisplatin-based chemotherapy for outpatient treatment in lung cancer and mesothelioma

Conclusions: These observations indicate that intermediate to high-dose cisplatin administration is feasible in outpatient management with a short hydration regimen without high risk of nephrotoxicity.

June 15th, 2007. Phase II study of erlotinib in patients with malignant pleural mesothelioma: a Southwest Oncology Group Study

Conclusion: Single-agent erlotinib was not effective in MPM, despite high expression of EGFR. Activation of the ERK and phosphatidylinositol 3-kinase/Akt downstream pathways are possible resistance mechanisms to EGFR TKI. The activated phosphatidylinositol 3-kinase/Akt pathway is a potential therapeutic target for MPM.

June 15th, 2007. Generation and characterization of an ascitogenic mesothelin-expressing tumor model

Conclusion: The data indicate that the WF-3 murine tumor model may potentially serve as a good model for understanding the molecular oncogenesis of peritoneal tumors. In addition, the preclinical model may potentially be useful for the development of diagnostic and therapeutic methods against intraperitoneal cancers.

June 15th, 2007. Effect of the TAT-RasGAP(317-326) peptide on apoptosis of human malignant mesothelioma cells and fibroblasts exposed to meso-tetra-hydroxyphenyl-chlorin and light

Conclusion: TAT-RasGAP317–326 selectively enhanced the effect of mTHPC and light delivery on H-meso-1 cells but not on fibroblasts. However, this effect was mTHPC dose-dependent and occurred only at a low sensitizer dose.

Conclusions: Small numbers and limited information on jobs, occupational agents, and lifestyle preclude attribution of observed increases to workplace exposures.

June 15th, 2007. Primary omental Gastrointestinal stromal tumor (GIST)

Conclusion: Our case demonstrated a weak immunohistochemical expression of c-kit (CD117) and a point mutation in PDGFRA exon 12 resulting in an Asp for Val561 substitution. Imatinib therapy as an adjuvant to complete resection has been carried out safely. Because of the rarity of primary omental GISTs, it is inevitable to analyze accumulating data from case reports for a better and more detailed understanding of primary omental GISTs.

June 15th, 2007. Asbestos exposure and differences in occurrence of peritoneal mesothelioma in the Netherlands and Sweden

Conclusion: The absence of a time trend in the incidence rate of peritoneal mesothelioma in the Netherlands and Sweden in the past 15 years may point at a more limited role of occupational exposure to asbestos in the etiology of peritoneal mesothelioma than for pleural mesothelioma, especially among women. The observed drop around 2000 in annual incidence of peritoneal mesothelioma among Swedish women indicates the presence in the past of a substantial misclassification with other tumours in the peritoneum.
